摘要:
从江苏省沿海滩涂国家丹顶鹤自然保护区盐碱土样中,分离得到一株果胶酸裂解酶产生菌ZQX8,经形态、生理生化特征和16SrDNA鉴定,初步鉴定为枯草芽孢杆菌.该株菌的最佳培养条件为37℃、pH6.5、0.1~0.25mol/LNaCl,但在pH7~9之间,0.5~1.0mol/LNaCl浓度下仍然能较好的生长,表现出一定的耐盐性和耐碱性.该菌分泌的胞外果胶酸裂解酶的最佳底物为果胶酸,在培养基中酶活达到4U/mL.该果胶酸裂解酶在40~60℃,pH8~10的碱性条件下酶活较高,在600mmol/LNaCl存在的条件下,也能保持较高的酶活,显示该酶具有一定的耐碱性和耐盐性.
Abstract:
One pectate lyase producing strain, ZQX8, was separated from the sa lina so il in Yancheng Na tionalGrus japonensis NatureReserve, Jiangsu Prov ince. Through m oda lity of bacteria, spec ia lty in physi o log ic and b iochem ica,l and 16S rDNA, w e identified the bacter ia to be Bacillus subtilis. The results show ed that the m ost optim al cu ltu re conditions of th is bacter ium was: incubating at 37 ℃ , pH 6.5, w ith 0. 1~ 0. 25 m ol/L N aC.l But incubating a t 0.5 ~ 1.0 m o l/L NaC l and pH 7~ 10, the stra in could still g row, and th is showed that the stra in m ight have endurity ag ainst sa lt and sa lka l.i The stra in cou ld secrete pecta te lyase w ith enzym e activ ity 4U /mL med ium. The optium substra te o f th is enzym ew as po lygalactu ron ic ac id and the optium conditions was 40~ 60℃ , pH 8~ 10. The activ ity o f the enzym e still rem a ined at the higher concentration of 600mmo l/L N aC.l The enzym em ight have both sa lt-resistant and sa lka li resistant property.
