摘要:
通过对番红花及其混淆品的rDNA ITS区序列进行PCR扩增、测序,并运用Clustal X、Mega 3.0等软件进行序列分析.结果表明番红花rDNA ITS区序列全长650 bp,GC百分比为60.3%,与其混淆品的rDNA ITS区序列存在着显著差异.另外,还设计出了鉴别番红花的位点特异性PCR引物,无需测序即可对番红花及其混淆品进行准确的分子鉴别.
Abstract:
The PCR p roducts of rDNA ITS regions of Crocus sativus L. and its adulterantswere sequenced and analyzed using Clustal X、Mega 3.0 software. The results showed that the full length of rDNA ITS re gions of C. sativus was 650 bp, and the percentage of GC contents was 60.3% , which disp layed the re markable distinction between rDNA ITS regions of C. sativus and its adulterants. In addition, a pair of al lele-specific PCR (AS - PCR) p rimerswere designed to authenticate C. sativus and its adulterants accu rately without sequencing.
