摘要:
以脲素为底物,对曲霉脲酶的动力学特性进行了研究.在不同pH(5.0,5.6,6.0,6.6,7.0,7.6,8.0,8.6,9.0)条件下测定脲酶的Km值和Vmax.结果表明,pH对脲酶的Vmax影响明显,在pH5.5至pH8.0之间Vmax呈钟形.在pH7.0的酶促反应速度最快,Km值不受pH值影响;不同的pH值(5.0,5.6,6.0,6.6,7.0,7.6,8.0,8.6,9.0)下脲酶的Km值和Vmax显示,在pH7.6时对底物的亲和力最大,为最佳pH,该脲酶在pH6.6到8.6时的稳定性较好.
Abstract:
The dynam ic property of asperg illus urinary enzyme was investigated w hen urea was used as substra te. The va lue ofKm and Vm ax w ere tested under different pH cond itions( 5. 0, 516, 6.0, 6.6, 7.0, 7.6, 8.0, 8. 6, 9.0). The results showed that the va lue o f pH c lear ly affected theVmax . Vm ax show ed be ll shape in the range of pH 5. 5 to pH 8.0. The enzym ic reaction rate reached m ax imum as pH = 7. 0. The value ofK m was not influenced by pH va lue. The v alues o fKm and Vm ax unde r d ifferent pH ( 5.0, 5.6, 6.0, 6. 6, 7. 0, 7. 6, 8. 0, 8.6, 9.0) showed tha t the affinity to the substrate reached m ax im um when pH = 7. 6 and thus becam e the optim um pH. Th is ur inary enzym e exh ibited good stability in the range o f pH 5. 5 to pH8. 0.
