摘要:
测定了无蹼壁虎、粗疣壁虎和中国壁虎共17个样品的COI基因部分片段,结合其他已知序列,对我国9种壁虎的DNA条码序列进行比较分析,计算种间及种内平均遗传距离,并构建系统发生树.结果显示所有DNA条码都为各物种所独有,不存在物种之间的共享.属内种间差异明显大于种内差异,且同一物种的不同个体均聚成高支持率的单系群,提示该条码在壁虎属物种中有很好的识别能力,可以用于物种鉴定.DNA条码还为粗疣壁虎和铅山壁虎的物种有效性提供了有力的分子证据.
Abstract:
W e determ ined partial sequence of the cytochrom e c ox idase subun it I gene ( CO I) fo r 17 spec im ens from G ek- k o sw inhonis, G. scabridus and G. chinensis. Comb in ing that from ano ther 6 geckos, DNA ba rcodes for 9 spec ies ofG ek- k o from Ch inaw ere com pared. Average K imura-2-param eter differences be tw een and w ithin spec ies w ere calculated and the phy logenetic treesw ere reconstruc ted. Our results showed that a ll DNA barcodes we re speciesspec ific, that is no se quence w as shared in tw o spec ies. D ivergence betw een spec ies w as g rea ter than that ofw ithin species, and ind iv idua ls o f the sam e species w ere clustered as high ly supported m onophy le tic g roup, ind ica ting the po tential o f DNA barcoding for spec ies identification in genus Gekko. DNA barcodes a lso prov ide m o lecular ev idence suppo rting the valid ity o f Gekko scabr idus and G ekko hokouensis.
