摘要:
探讨吡咯喹啉醌(PQQ)对LPS诱导的巨噬细胞炎症因子基因表达的影响及机制. 培养RAW264.7细胞、BMDM细胞和THP-1细胞,分为对照组和实验处理组,用MTT方法检测PQQ对细胞增殖的影响,并分别用RT-qPCR和Western Blot检测各炎症因子的基因及蛋白表达情况. PQQ预处理能使LPS诱导的炎症因子表达量下降,并使P-P65蛋白表达下降,Nrf2蛋白表达增加. PQQ预处理降低细胞内炎症反应,这一过程可能通过增加Nrf2表达和抑制NF-κB表达而发挥作用.
Abstract:
To investigate the effects of pyrroloquinoline quinone(PQQ)on the expression of inflammatory factors in LPS-induced of macrophages. Cultured RAW264.7 cells,BMDM cells and THP-1 cells were divided into the control and PQQ-treated groups,respectively. MTT method was used to detect the cell proliferation. The expression of each inflammatory factor at mRNA and protein levels was detected by RT-qPCR and Western Blot analysis. PQQ pretreatment caused a significant decrease in the expression of inflammatory factors,the expression of P-P65 protein and consequently an increase in the expression of Nrf2 protein. PQQ pretreatment reduces the intracellular inflammatory response,which may attribute to an increase in the expression of Nrf2 and resultant inhibition of the expression of NF-κB.
