Abstract:
Plasmamembrane(PM)depolarizationcanenhancetheproliferationofcertaintypesofcellsincludingsatellitecell.However,theeffectsofPMdepolarizationontheproliferationofmyoblasthavenotbeendetermined.Inthisstudy,itwasinvestigatedtheeffectsofPMdepolarizationviahighconcentrationofextracellularpotassiumion(K+)ontheproliferationofmurineprimarymyoblasts.Bydirectcellcounting,itwasshownanapparentincreaseinthenumberofcellsculturedinmediumcontaining25mmol/LK+comparedwiththatofregularmedium(5mmol/LK+).However,excessivelyhigher(50mmol/Land100mmol/L)concentrationsofK+didnotpromotethegrowthofmyoblasts.Inaddition,adecreaseinthenumberofcellsundergoingG1phaseandanincreaseinthatofSphasewasdetectedbyFACStechnology.ImmunostainingandWesternblotanalysisshowedthatthemyogenicdifferentiationpotentialwasnotalteredafterPMdepolarizationtreatment.Furthermore,itwasfoundthatthebasallevelofactivatedextracellularsignal-regulatedkinase1/2(ERK1/2)wasenhancedwithPMdepolarizationtreatment.BothU0126andPD98059,theMEK1/ERK1/2inhibitors,obviouslyattenuatedtheproliferationenhancementofthecells.Takentogether,ourdataindicatethatPMdepolarizationpromotestheproliferationofprimarymyoblaststhroughactivationofMAPK/ERK1/2pathway.
