摘要:
采用Qiagen的植物基因组DNA提取试剂盒并加以改进,首次从不同季节采收、不同干燥方法保存的三种罗布麻(罗布红麻、大叶白麻和白麻)干燥叶片中提取出基因组DNA.提取的基因组TE溶解液的OD260/OD280值多在1.7-1.9范围内,电泳呈现单条带,核基因组的ITS片段、叶绿体基因组的trnL内含子和trnL-F非编码区片段的PCR扩增条带清晰并能进行DNA测序,这表明基因组DNA质量较好,可以满足后续分子生物学实验的要求.本研究为罗布麻等中药材的进一步分子生物学研究提供了必要的技术基础.
Abstract:
Genom ic DNA w as first iso lated from dr ied leaves of Luobum a by the mod ified m e thod of the Q iagen DNeasy plant m in i k it. Resu lts showed tha t by th is me thod genom ic DNA cou ld be iso lated from Luobum a leaves o f three species be ing co llected at d ifferent seasons and be ing preserved by d ifferent m ethods. The OD260 /OD280 ratio s of the iso lated DNA w ere in the range o f 1.7~ 1.9, and only one band presented in agarose ge ls; c lear PCR bands o f ITS rDNA and trnL intron, trnL- F non- cod ing cpDNA cou ld be sequenced.
