摘要:
NRAGE基因作为MAGE家族的一员,在细胞周期、细胞增殖和细胞凋亡过程中均发挥着重要的作用.关于NRAGE对神经元细胞分化的影响目前虽有两篇文献报道,但结论相反,且两篇文章均是以大鼠肾上腺髓质嗜铬瘤PC12细胞为模型进行的实验.在本文中,我们分离了NRAGE基因敲除小鼠的海马,以新生大鼠神经胶质细胞为饲养层,通过原代培养得到了NRAGE完全敲除的神经元细胞,通过统计神经元细胞的轴突/胞体直径比例和树突数目,分析了NRAGE基因敲除对神经元细胞分化的影响.实验结果表明,野生型和敲除型神经元细胞的分化没有发现明显的统计学差异,提示NRAGE基因对小鼠神经元细胞分化没有影响.
Abstract:
NRAGE,whichbelongstoMAGEfamily,playsanimportantroleinthecellcycle,proliferationandapoptosis.TheinfluenceofNRAGEondifferentiationofneuronshadbeenreportedintwopaperswhichindicatedtwooppositeconclusions.ThesetwoarticlesbasedonPC12cells,whichderivedfromapheochromocytomaoftheratadrenalmedulla.OurexperimentisolatedhippocampalsofNRAGEknockoutmiceandtookneonatalratglialcellsasfeederlayertoobtainfetalmiceprimaryneurons.DetectedtheinfluenceofNRAGEgeneonneuronsdifferentiationbycountingneurite/diameterofsomaandthenumbersofdendrites.Theresultsshowedthatprimaryneuronaldifferentiationsofwild-typeandknockoutfetalmicehadnosignificantdifference,indicatingNRAGEgenehasnoeffectontheneuronsofmice.
