摘要:
通过溶液培养试验,研究了不同Mn 2+浓度(0、0.09、0.18、0.27、0.36mmol/L)胁迫对菹草抗氧化系统、活性氧产生和亚显微结构的影响.结果表明:随着Mn 2+浓度的增加,菹草叶片中的叶绿素a、b和总叶绿素含量都呈现先升后降的趋势;超氧化物歧化酶(SOD)活性逐渐上升,而过氧化物酶(POD)和过氧化氢酶(CAT)活性则呈先升后降趋势,还原型谷胱甘肽(GSH)和抗坏血酸(AsA)含量呈先升后降趋势,同时超氧阴离子(O.2-)产生速率、过氧化氢(H2O2)、丙二醛(MDA)含量均呈逐渐上升趋势;电镜观察发现, M n 2+ 对叶片细胞器的亚显微结构特别是叶绿体、线粒体和细胞核造成严重损伤.M n 2+ 破坏了菹草正常生理活动的结构基础和离子平衡, 并造成抗氧化系统功能紊乱, 这些都是M n 2+ 对菹草产生毒害的重要原因.
Abstract:
The tox ic effect of various concentrations ofMn 2+ ( 0、0. 09、0. 18、0. 27、0. 36mmo l/L) w as stud ied on antiox-i dant system, active oxygen generation and subm icro scop ic structu re of Po tam og eton crisp us. The results show ed that the content of chlorophy ll a, ch lo rophyll b and to tal ch lorophyll a ll first increased and then decreased w ith increas ing the concentration o fM n 2+  . Superox ide d ism utase ( SOD ) activ ity g enerally increased, peroxydase ( POD ) and catalase ( CAT) ac tiv ities enhanced a several fo ld, but w ith the m anganese concentration increased continuously, they a ll dec reased, wh ich w as po ssib ly the reason o f accumu lation o f O- 2 and H2O 2. G lutathione ( GSH ) and antiscorb ic ac id ( A sA) conten ts changed at the sam e trend o f POD and CAT. The leve l o f lipid perox idation w asm easured as the content o fm a londialdehyde (MDA), w hich w as a lw ays increased and w as the same as the change o f O- 2 and H  2O 2. Transm iss ion electron m icroscopy observation indicates that the h igherM n2+ concentration, the mo re serious dam ages to the leaf ce lls, especia lly ch lo roplast, m itochondrion and nucleus. The conclus ion could be reached tha t the grow th declin ing even death of plantw as resu lted from destruc tion o f structure foundation of physiolog ica l function, interruption o f ion and enzym es equilibr ium and disorde r o f an tiox idant system. But the influences of those aspects a re in different ex ten t in Po tam ogeton crispus.
